簡介背景 (研究建構+研究問題) Commonly found in legume seeds, proteinase inhibitors play a critical role in regulating several physiological processes such as blood coagulation, complement fixation, fibrinolysis, and fertilization. As proteins having characteristic polypeptide scaffolds, most of these inhibitors are grouped into several families including the Kunitz, Kazal, Burman Birk, Serpin and mucus families. As is well established, the production, invasion and transfer of cancer cells are significantly related to proteinase inhibitors. However, previous efforts have failed to suppress cancer proteinase inhibitors effectively, largely owing to the inability to identify proteinase inhibitors that can suppress more than 80﹪of all cancer cells. 目標 Therefore, this study attempts to purify and characterize the coffee senna trypsin inhibitor (CSTI), to which Coffee Senna, i.e. a leguminosae that belongs to its bean sub-family, by using protein purification. Essential to understanding the fundamental principles of protein purification, protease inhibitors are used to design new substances, capable of effectively controlling diseases and pathological processes. 方法 Coffee senna seeds are analyzed. CSTI is then extracted using a crude extraction method containing ammonium sulfate precipitate. Next, the senna trypsin inhibitor is purified to utilize the column (i.e., gel filtration, affinity chromatography, and ion exchange column) of various characteristics (i.e. molecular size, molecular charge, as well as molecular polar and affinity). Additionally, the purified protein is analyzed to further characterize its heat stability and different SDS concentrations in order to achieve stability. 歸納希望的結果及 Analysis results demonstrate the feasibility of extracting CSTI and identifying the type of coffee senna seed. Besides characterizing CSTI precisely through analysis, its extraction and characterization are used to classify the coffee senna type. Moreover, the CSTI sequence achieves sequential alignment of the protein with other legumes. 其對特定領域的貢獻 In addition to providing insight into the physiological significance of CSTI in plants, results of this study contribute to efforts to design substances capable of effectively controlling diseases and pathological processes.

商用提案建構 While facing an increasing elderly population, Taiwanese must effectively address the increasing demand for long-term healthcare facilities and services. The potential growth for this non-profit market sector, the largest one in Taiwan, is immense, making it impossible for healthcare providers to resist expansion into this area. Both governmental policymakers of social welfare trends and commercial investors heavily rely on forecasting reports to remain abreast of regulations governing health finance policies and to develop new inventory projects. 商用問題 Whereas the modeling method has seldom been adopted to forecast market trends in long-term care in Taiwan, most studies focus mainly on the expert conjecture method or the principle of the past growth relative proportion to forecast the market supply and demand. Those methods not only neglect the market and the natural factors that are correlated to the long-term healthcare sector, but also resulted in individual and organizational estimates in which a high degree of variance exists for the demand. 商用問題的量化 The World Health Organization (WHO) defines an aging society as one in which 7% of a country’s population is over 65 years old. Under this definition, Taiwan became an aging society in September 1993, with its elderly population reaching 1,470,000. Moreover, the island’s growth rate of the aging population exceeds that of European and North American countries. 問題的中心 Such a high degree of variance makes it extremely difficult for institutional managers as well as public and private sector investors not only to determine the accuracy of forecasting reports, but also to determine what policies or inventory projects to implement. Those managers and investors remain wary of committing to this market sector owing to the high degree of unforeseeable risks. 商用計劃需求 Therefore, two forecasting models must be developed to estimate the market demand of the elderly population in Taiwan with respect to the available resources in the long-term healthcare sector.
